Novatio and his brother, the martyr Timothus, were sons of Saint Pudens and Claudia Rufina and brothers of Saints Pudenda and Praxedes, their paternal grandfather was Quintus Cornelius Pudens, the Roman senator, who with his wife Priscilla They were among the first converts of St. Peter in Rome and in whose house they dwelt while they were in that city. It is believed that part of the modern structure of the church of Santa Pudenziana (Vía Urbana) is part of the senatorial palace or of the baths built by Novacio.

According to the fifth-century historian Filostorgio, Novacio was of Phrygian origin.
